Persistent pain is a common and significant medical condition with diverse impacts on people’s lives. Codeine phosphate is a regularly prescribed analgesic for moderate to severe pain, and is reliant on biotransformation to morphine by the CYP2D6 enzyme for efficacy. In this study, we investigated the codeine response in Caucasians with persistent pain using CYP2D6 genotyping and urinary metabolite concentrations to develop a biomedical model for predicting long-term codeine responses. This model could aid clinical decision-making when prescribing codeine and improve analgesic responses.
